Abstract: Brown spot disease is one of the primary factors afecting safety of leaf tobaccoquality in Changsha tobacco-growing region.Toinvestigate theocurenceandidentifythepathogenicspeciescausing tobacobrownspotdisease,acomprehensivesurvey wasconductedonincidencerateand diseaseindexofbrownspot diseaseacrossthe tobacco production towns in Changsha.Results showed that the incidence rate and disease index ranged 0 . 5 6 % - 1 6 . 6 7 % and 0.06-5.43 respectively. Through combined morphological andmolecularbiologicalidentificationofsolatedpathogens,thestudyrevealedthatthecausativeagentsereAlternariaalteata and A. tenuissima, with the detection rate of A .tenuissima up to 7 5 . 0 0 % . Further in this study, rDNA-ITS gene was confirmed to be excesive conservationforspecies-leveldiferentiation withintheAlternariagenus,whereastheplasma membraneATPasegene displayedconcordance withmorphologicalidentificationresults,provingsuitableforinterspeciesdiscriminationInconclusion,the pathogensoftobaccobrownspotdiseaseinChangshatobacco-growingregionexhibithighdiversity,withAlternariaalernatabeing predominant.
