中等強度耐力運動對老年性骨質(zhì)疏松癥小鼠NLRP3/Caspase-1/GSDMD焦亡通路的影響

[Abstract]ObjectiveToinvestigatetheefectsof moderate-intensityenduranceexerciseonsenileosteoporosis (SOP) throughtheNOD-likereceptor family pyrindomaincontainingprotein3 (NLRP3)/cysteineprotease-1 (Caspase-1)gasdermin D (GSDMD)signaling pathway.Methods Twenty-one18-month-old maleC57mice wererandomlydivided into three groups:the18- month-oldgroup,the2O-month-oldgroup,andtheexercisegroup,with seven mice ineach group.The SOPanimal model was replicatedusing naturallaged malemice,andanadditional seven six-month-old maleC57mice wereselectedas theyounggroup. Afteroneweek ofadaptivefeding,miceintheyoung groupand18-month-oldgroupwereeuthanized forsamplecolection.Mice intheexercisegroupunderwentmoderate-intensityendurancetraining,whilethoseinthe2O-month-oldgroupreceivedno intervention.Samples werecoletedaftereightweeksof intervention.Bone mineral density(BMD)of theleftfemur ineachgroup of micewasmeasuredusing micro-computedtomography(Micro-CT.ELISAwasused todeterminetheserum levelsof bone metabolismmarkers,ncludingC-terminaltelopeptideoftypeIcollgen(CTX-1),tartrateresistantacidphosphatase(AC) terminal propeptdeoftyeIprocollgen(INP),andbospcificalalinephospatase(B-APInditin,levelsofitein 1β(IL-1β)andinterleukin-18(IL-18)intherightfemurwereasessedWesternblotanalysiswasconductedtodeteminethe proteinexpresionlevelsofNRP3,Caspase-1,andtheN-terminalfragmentofGSDMD-Nintherightfemurofeachgroup. Results Compared with theyoung group,the18-month-oldand2O-month-old groups showed increased serum levelsof CTX-1and TRACP,as well as elevated expression levels of NLRP3,Caspase-1,GSDMD-N, IL- 1β ,andIL-18intherightfemur( P <0.05, P< 0.01).Meanwhile,serum levels of PINP and B-ALP,along with BMDof the left femur,decreased ( P <0.05, P?0.01 1).Compared with the18-month-oldgroup,the2O-month-old group exhibited increases inserum CTX-1andTRACPlevels,aswellasinNLRP3, Caspase-1, GSDMD-N, IL-1β,and IL-18 expression levels in the right femur ( P?0.05 P?0.01 ),along with decreases in serum PINP and B-ALP levels and left femur BMD ( P <0.05, P <0.01).Compared with the 2O-month-old group, the exercise group showed reductions inerumCTX-1andRACPlevels,aswellasinrightfemoralexpresionlevelsofLRP3,Caspase-1,GD-N,IL 1β,and IL-18 ( P <0.05, P
