卡瑞利珠單抗聯(lián)合順鉑誘導(dǎo)食管鱗癌Eca-109細(xì)胞凋亡的研究

Abstract:Inorder to study the effctofcarilizumabcombined with cisplatinonapoptosis of esophagealsquamous cell carcinoma celsEca-109byregulating mitochondrialoxidative phosphorylation (OXPHOS)andphosphatidylinositol3 kiase/ proteinkinaseB(PI3K/AKT)signaling pathway,andtoobservetheexpresionofheatshock proteinA9(HSPA9).Inthis study, the test was divided into blank control group,cisplatin group (100μmol/L )and combined administration group which is cisplatin (100μmol/L)? +carrilizumab 200mg/time ).Transwell migration test,scratch test,cell counting kit8(CCK-8) test andclonalaggregation technique were used toobserve the effectsofcarrelizumabcombined with cisplatinontheinvasion, migrationand proliferationof esophagealsquamouscellcarcinomacelsEca-19.The expressionofHSPA9gene wasdetected by RT-PCR. Westernblot(WB)was used to detect the expresion ofOXPHOS and PI3K/AKTpathway proteins.Mitochondrial reactiveoxygenspecies (ROS)were detected byflowcytometry.Transwellmigration,scratchtest,CCK-8 testandclonal formationtestshowedthatthecombinedadministrationgroupihbitedtheactivitymigrationandaggregationofEca-O9cells more than the cisplatin group.Reverse transcription-polymerase chain reaction (RT-PCR)and WB detectionresults showed that compared with cisplatin group and control group,the expresion of HSPA9 and OXPHOS in combined administration group significantly decreased,and the phosphorylation of PI3K/AKTwas inhibited (P<0.01 ). Flow cytometry showed that compared withcontrolgroupandcisplatin group,ROSlevelsincisplatin+carilizumab groupsignificantly increased,andtherewas a significantdifference ?P<0.05 ).Compared with cisplatinalone,carrellizumabcombined with cisplatincould significantly inhibit the expressionofHSPA9andOXPHOS inesophagealsquamous cellcancercells,ihibit thephosphorylationofPI3K/ AKTsignalingpathwayandregulatereoxidationandmitochondrialhomeostasis.TheinhibitionofEca-1O9onesophageal squamous cellcarcinoma by inducing mitochondrial apoptosis provides a reference forthe precise interventionandtreatmentof esophageal squamous cell carcinoma in clinical practice.

食管癌(esophagealcancer)是臨床上發(fā)病率較高、惡性程度較高的一類消化道腫瘤,因臨床癥狀較隱匿,早期不容易被及時(shí)發(fā)現(xiàn),等出現(xiàn)臨床癥狀時(shí)已是中晚期。(剩余12817字)
