瑪卡總皂苷促人視網(wǎng)膜母細(xì)胞瘤Y79細(xì)胞凋亡作用研究

AbstractTo investigate the effct and mechanism of Lepidium meyenii Walp.saponin (LMS) in promoting apoptosis ofhumanretinoblastoma Y79cels,Lepidium meyenii Walp.wasusedas theexperimentalmaterial to extract its saponins.Y79 cells were treated with different concentrations of LMS (O,10O,2OO,400,and 8 0 0 μ g / m L ),and the effcts ofLMSonY79cellproliferationandapoptosisatdiffrent treatmenttimes (24,48,and72h)weredetected.The expression levels of pro-apoptotic factors, including Caspase , Caspase-8, and Caspase genes and proteins, were measured.The results showed that 2.58 g of total saponins were extracted from 5O g of maca dry powder,with an extraction yield of 5 . 1 6 % . LMS treatment inhibited the proliferation of Y79 cells and induced their apoptosis,with the optimal effect observed at 4 0 0 μ g / m L for 48 h,resulting in a cell viability rate of 5 4 . 0 5 % and an apoptosis rate of 2 1 . 8 7 % .Under these conditions,LMS promoted the expression of Caspase-3,Caspase ,and Caspase-9 genes and upregulated the expression of Cleaved caspase , Cleaved caspase-8,and Cleaved caspase proteins.In conclusion, LMS can inhibit theproliferation of Y79cellsand induce their apoptosis,and its mechanism maybe related to the upregulation of pro-apoptotic factors Caspase ,Caspase-8, and Caspase
