Abstract:【Objective】Chlorogenicacid isanindex componentforthequalityevaluationofEucommiaulmoides leaves,whichhas variousmeiciallusschtbactealdtamtorytiodanttacrdtptitieatio andexcaationofmetabolicpathwaysandgnesinvolvedinregulatingchlorogenicacidbiosynthesis inEucommiaulmoidesleaves willprovidefurtherevidenceforanalysisofthemolecularmchanismofchlorogenicacidformationinEucommiaulmoidesleaves. 【Method】Eucommialmoidesleavesrecectedfromotou,Iergoliautonomousgion,inpril,Julydctober 2023at diferetdevelopmenalperods,andwereclaifedaccordingtotecharacterationas:Aprilfreshlyunfoledyoungleaves; Julymatureleaves,withlighteathery;andctoberfullymatureaves,withthdarkestcoloroftelavesandompleteleathrye chlorogenicacidcontentofEucommiaulmoidesleavesatdiferentperiodswasdeterminedbyig-performanceliquidchromatography (HPLC),andthediferetiallexpressedgenesereidentifdbyrasiptomesequencing.【Result】Teloogniccidctet ofEucommiaulmoidesleavesrangesfromApriltoJulyandthentoOctober,showsanincreasingandthendecreasing trend withleaf development.Transcriptome sequencing yielded 58.06 Gbof clean data,enablingthe identification of11 689 genes.The numberof down-regulatedexpressdgnesisigherthanteumberofupregatedexpresedgenesinteompasonofsTTsand
T2vsT3hreegroups.KEGGenrichmentanalysisandGOenrichmentanalysis,bothofwhichdisplaychlorogenicacidsynthsis-related enrichments.FurtemoreKmeanstredexpreionividsteensinto0modles,inwhichtrendsofgeneexpressinlass4, Class8adClassareconsstentwithtrendsoforogenicacidotetadtwozsandegensareidentifdfroClass4 Class8andClass10tatarerelatedtoclorogenicacidbiosythsis.Tresultsofal-tiefuoreseequanitativePCR(-CR) are inaccordance withRNA-seqoutcomes.【Conclusion】ThechlorogenicacidcontentintheEucommiaulmoides leavesshowsa trend of increasing and then decreasing with the development of the leaves.
