Expressionandclinical significance ofKLF4 inplacenta and serum ofpatients withpreeclampsia

[Abstract]Objective：To explore therelationshipbetween Kruppel-like factor 4（KLF4）andthe developmentof preclampsia（PE），anditsclinical significance.Methods：Atotalof68patients withPEwereselected，including37cases of early-onset preeclampsia （EPE） （EPE group）and 31 cases of late-onset preclampsia （LPE）（LPE group）.Sixty normal pregnant women inthe third trimester of pregnancy were taken as theNP group.Theimmunohistochemistryand protein bloting method wereused todetect the expresionof KLF4in placentaltissues;theenzyme-linked immunosorbentassay （ELISA）wasusedtodetectthelevelofKLF4inperipheralbloodandumbilicalarteryblood，andPearsoncorrelationtest wasused to analyse the relationship between serum KLF4 level inthe patients with PEandthequantitative amount of 24 h urineprotein.Results：TherelativeexpressionlevelofKLF4 protein inplacentaltissuesof thePE group wasO.61±0.10， which waslower than O.98±0.11 of the NP group（ P <0.05）; the relative expression level of KLF4 protein in placental tissues of the EPE group was 0.34±0.09，which was lower than 0.67±0.12 of the LPE group（P<0.05）.The concentration of KLF4 in the peripheral'serum of the PE group was 1.78±0.09 ，which was lower than 1.85±0.08 of the NP group（ P< 0.05）；theconcentration of KLF4intheumbilicalarteryserum of thePE groupwas1.56±O.13，which was lower than 1.66±0.08 of the NP group （P<0.01） ； the correlation analysis showed a negative correlation between the level of KLF4 in the PE group and the quantitative amount of 24 h urine protein （r=-0.7150， P <0.001）.Conclusions： The expression of KLF4 isdecreased inplacental tisues，peripheralbloodandumbilicalartery bloodof PEpatients.Thelevelof KLF4 in peripheral serum isnegativelycorrelated withthe24hurine proteincontent，suggesting thatthelowexpressionofKLF4is related to the occurrence and development of preeclampsia.
