Abstract:Quercetinisaflavonoid compoundwithvarious biological activities,mostly existing inthe formof glycosides in plants.Sophora japonicais an idealraw material for preparingquercetin,with advantagessuch as wide plantingarea andlow price.Thisstudy investigated the synergistic antibacterial effects ofquercetin against Salmonella spp.thatcombined with colistin.Thequercetin wasprepared fromthe Sophora japonica buds Aspergilus niger fermentation and isolated from thesemi-solid fermentation productsusing ultrasonic extraction methods, purified and structurallyidentified using HPLC,ESI-MSandNMRmethods.Theantibacterialactivitiesofquercetin andcolistin,both individuallyandincombination,against Salmonella were evaluatedusing dilutionand checkerboard methods.The results showed that the highest product of quercetin was 73.7 mg. g-1 after fermentation for 96h and the purity reached to 98 % afterpurificationand identification.After evaluation of antibacterialactivity,the minimum inhibitoryconcentrations(MICs)of colistin against S.typhimurium,S.pullrum 1789 and 520 were 2,8, and 4μg?mL-1 ,respectively,while the MICs of quercetin against the mentioned strains were all above 500μg?mL-1 When quercetin wasused together with colistin,the synergistic antibacterial effects were observed against above
Salmonella strains,particularly against S.pullrum1789and 52O,with fractional inhibitoryconcentration index (FICI)valuesbelow O.5,and the MIC value of colistin were reduced to 1μg?mL-1 . The study demonstrated the synergisticinhibitoryefectofquercetinpreparedfrom Sophorajaponica throughA.nigerfermentationcombinedwith colistinagainst Salmonella,providing ascientific basis fordeveloping newantibacterialcombination strategies.
