AbstractDaning sun-cured tobacco is a species of good-quality yelow sun-cured tobacco in Hezhou， Guangxi，and it is the excellent raw material for cigarette production.However，it has extremely low resistance to black shank disease.This study investigated the response mechanism of Daning sun-cured tobacco to black shank disease by comparing the diferentially expressed genes and metabolites between healthy and black shank-infected leaves through transcriptomic and metabolomic analyses.The results showed that a total of 7 194 significantly diferentially expressed genes were identified through transcriptomic analysis，and 198 significantly diffrent metabolites were detected through metabolomic analysis.The combined transcriptomic and metabolomic analysis revealed that ： ① Plant hormone signal transduction and phenylpropanoid biosynthesis pathways played important roles in the defense of Daning sun-cured tobacco against black shank infection. ② 二 was speculated that metabolic pathways such as aminoacyl-tRNA biosynthesis，cyanogenic amino acid metabolism，valine，leucine and isoleucine degradation，cysteine and methionine metabolism，and arginine and proline metabolism were related to the response of Daning sun-cured tobacco to black shank stress.This study elucidated the major gene transcriptional regulatory networks and metabolic pathways of Daning sun-cured tobacco in response to black shank infection，which could provide a theoretical basis for research on improving the resistance of Daning sun-cured tobacco to black shank disease.
